"Mission to the Unknown" was a regular episode of Doctor Who, produced and broadcast as a prelude to "The Daleks' Master Plan". It was not the first spin-off, but it was the first "Doctor-lite" story (indeed, the Doctor doesn't appear at all). Terry Nation, who wrote the episode, later took some of the characters from Master Plan and attempted to launch a US-produced spin-off series featuring the Daleks, but nothing came of it beyond a few scripts.
